Bacteria have plenty of ways to combat the viruses that plague them, called phages. The CRISPR-Cas system and restriction enzymes that cleave phage DNA are the best understood, but there are others. They use a variety of different mechanisms and stop phages at various stages of infection, but they all rely on biological actors: proteins and RNA. New work has just revealed, though, that bacteria can use chemical weapons as well.
Bacteria produce a wide range of active small molecules that are not essential for survival but do confer a growth advantage. Some of these small molecules, which kill their fellow microbes, are already used (by us) as antibiotics. It was also observed, more than fifty years ago, that bacteria make molecules that can inhibit the growth of phages. But it wasn’t clear whether these molecules are made specifically because they slow down the phages. Only now, with the background knowledge that (a) bacteria make a lot of bioactive compounds, many of which do combat other microorganisms, and that (b) phages are a major scourge for bacteria, did researchers think to check.
Biochemists screened 4,960 compounds for their ability to protect E. coli from phage infection and found 11 that could. Nine out of the 11 were what are termed DNA-intercalating agents. The nucleotides that comprise DNA (A,T, C, and G) are flat molecules, and they’re stacked parallel to each other along the DNA helix. DNA-intercalating agents slide in between them, interfering with the copying of DNA during cell division.
]]>The network behind ‘Game of Thrones’ has reportedly partnered with Carnival Films, the production company behind ‘Downton Abbey,’ to adapt a 2016 documentary by Alex Gibney. According to Deadline, Gibney himself will direct the project, which will reportedly also involve ‘The Americans’ screenwriter Stephen Schiff.
“I first came across it when I was doing We Steal Secrets,” the film about Julian Assange and Chelsea Manning, Gibney told Deadline. “I didn’t understand at the time that it was something deeply fundamental about where we were heading, in terms of spying and cyber conflict. That didn’t happen until we started to make the film.”
Gibney credited Marc Shmuger, who produced ‘Zero Days,’ with opening his eyes to the pivotal importance of Stuxnet. The virus “set in motion a new kind of arms race,” he said.
“It sent another kind of message, too, which is, the United States and Israel will use weapons and attack people first, and that sets a different kind of precedent for other countries, as well: Why shouldn’t we do the same?” Gibney told Deadline. “You can say that what we did with Stuxnet was an undeclared act of war — it was an attack on critical infrastructure in a time of peace. That sets a terrible legal precedent.”
First detected in 2010 by Russia’s Kaspersky Labs, Stuxnet is widely believed to be the work of the US and Israel. NSA whistleblower Edward Snowden confirmed that claim in a 2013 interview. However, neither government has officially acknowledged being behind the virus.
Computer code found in the 2017 leak from Shadow Brokers, which was allegedly stolen from the NSA, was also found to match that used in Stuxnet. Designed to target industrial control systems used in infrastructure facilities, Stuxnet modifies data on controller software that affects their automated processes.
In 2011, Iran accused the US and Israel of employing Stuxnet to attack Tehran’s nuclear program, specifically the uranium enrichment facility at Natanz. Stuxnet was reportedly introduced into the facility on a USB flash drive, bypassing the plant’s isolation from the internet.

Plumbing the depths of women’s bladders may shower researchers with viral gold.
In a wee survey, Loyola University Chicago researchers found the sac-like organ brimming with never-before-seen viruses that can kill and manipulate bacteria. Their findings, published this week in the Journal of Bacteriology, offer a first-pass catalogue of the rich diversity of bacteria-infecting viruses—aka “phages” or “bacteriophages”—in the bladder microbiome. The researchers suggest that further studies into the streaming viral content could one day lead to phage-based methods to void bacterial infections and identify disorders.
“The thought that there’s not bacteria in urine is false,” Catherine Putonti told Ars straight away. Putonti, a bioinformatics researcher and microbiologist at Loyola, is the leading author of the study. “The big picture is that there are a lot of viruses that are part of these bacterial communities as well.”

Could an alien virus arrive from outer space and wipe out the human race? It may sound like the plot of a B-movie, but an international group of scientists is now formally exploring the possibility for the very first time.
In a study published in the journal Astrobiology next month, the group sets out its belief that while NASA continues to focus its search for extraterrestrial life on the hunt for other alien microorganisms, space viruses remain critically under-researched. The team, led by biologist Kenneth Stedman of Portland State University, is calling for scientists to develop strategies to detect these viruses – and to see if they can spread to humans from outer space.
“More than a century has passed since the discovery of the first viruses,” Stedman said in a statement. “Entering the second century of virology, we can finally start focusing beyond our own planet.”
Viruses are infectious agents that replicate inside the living cells of other organisms. They can infect anything, from animals and plants to microorganisms like bacteria. The team behind the study believes that because more viruses exist on Earth than any other cellular organism, they should exist on other planets, too. The pursuit to find these astronomical agents has led Steadman, along with colleagues Aaron Berliner and Tomohiro Mochizuki from UC Berkeley and Tokyo Institute of Technology respectively, to create a new discipline in astrobiology known as “astrovirology.”
“With this paper, we hope to inspire integration of virus research into astrobiology and also point out pressing unanswered questions in astrovirology, particularly regarding the detection of virus biosignatures and whether viruses could be spread extraterrestrially,” Stedman said.
Speaking to Gizmodo, Stedman tried to allay fears about the future discovery of viruses in outer space. “Viruses have a bad rap. If we find viruses on other planets it is an indication of life, not something to be scared of,” he said.

For the past 3 years, the US has maintained a moratorium on backing research that involves genetically modifying viruses to make them more potent, whether it’s their ability to spread or their lethality. You can kiss that de facto ban goodbye, however: the government has lifted the restriction in favor of a new review step. Scientists who want to engineer these super viruses will be subject to scrutiny by a “multidisciplinary group” that will consider the intentions and risks, such as whether or not it’s “ethically justifiable” and whether or not there are safer methods of accomplishing the same goal.
If you ask officials, this shouldn’t amount to a serious change. The National Institutes of Health’s Dr. Francis Collins said that 10 of the 21 studies frozen by the moratorium went ahead anyway thanks to exemptions, and this is simply a matter of “regularizing the process” of making those exemptions.
That isn’t making some researchers any less nervous. There are certainly benefits to the move: creating these mutant viruses could help by anticipating their evolution and developing treatments before they become especially dangerous. And in some ways, gene modification is necessary to see how viruses change based on their genetic structure. The perils, however, are obvious. These gene-modified viruses could wreak havoc if they escape the lab, and there have been mishandling incidents for everything from avian flu to Ebola.
In that sense, the concern isn’t so much lifting the moratorium as it is human nature. Even the most diligent biologist might not spot a tiny leak in a biohazard suit, and some viruses may not produce signs of infection until it’s already too late. Scientists will have to think very carefully before making proposals, and they’ll likely want to take extra precautions to avoid even the hint that a super virus might get free.

A team of Swiss scientists have created artificial viruses that stimulate the immune system which in turn activates and sends a “powerful army” of so-called killer cells to fight cancerous tumors.
Cancer cells are notoriously good at evading the immune system, but viral infections cause the body to act quickly, stimulating the system to use all available means to fight the unwanted invader.
This new designer therapy capitalizes on the body’s ability to quickly spot a virus, so that when the immune system “sees” it, it can hunt down cancer, hopefully killing the tumor, according to the press release published by the University of Basel.
Researchers, lead by Professor Daniel Pinschewer from the University of Basel and Professor Doron Merkler from the University of Geneva, built artificial viruses based on lymphocytic choriomeningitis virus (LCMV), which can infect both rodents and humans.
Although the viruses were not harmful for mice, they did produce alarm signals which were picked up by the human immune system. The team of virologists also integrated proteins found in cancer cells into the viruses.
Infected with the harmless designer virus, it enabled the immune system to recognize these cancer proteins as dangerous, allowing the body to create a “powerful army of cytotoxic T-lymphocytes, also known as killer cells, which identified the cancer cells through their protein and successfully destroyed them.”
“We hope that our new findings and technologies will soon be used in cancer treatments and so help to further increase their success rates,” the study’s senior author, Professor Pinschewer said in a statement.
The team’s research was published Saturday in the Nature Communications journal.
