Proteins are chains of amino acids, and each link in the chain can hold any one of the 20 amino acids that life relies on. If you were to pick each link at random, the number of possible proteins ends up reaching astronomical levels pretty fast.

So how does life ever end up evolving entirely new genes? One lab has been answering that question by making its own proteins from scratch.

Way back in 2016, the same lab figured out that new, random proteins can perform essential functions. And those new proteins were really new. They were generated by scientists who made amino acid sequences at random and then kept any that folded into the stable helical structures commonly found in proteins. These proteins were then screened to see if any could rescue E. coli that were missing a gene essential to survival.

The single most abundant protein on the planet isn’t actually very good at its job. And, unfortunately, its job is important: to pull carbon dioxide out of the atmosphere and incorporate it into sugars and other molecules that most of Earth’s life depends on. Improving its function could help us in a variety of ways, from boosting crop productivity to cleaning up after our carbon emissions.

Unfortunately, the enzyme is also extremely fussy about how it operates, in part as a result of the evolutionary events that put it in plants in the first place. But now, a team of German scientists has figured out how to get the enzyme to work in the standard lab bacteria, E. coli, opening the door slightly to genetically engineering our way to more efficient plants. But the work also makes it clear that things aren’t quite as simple as we’d like.

A key enzyme

The enzyme has the catchy name “ribulose-1,5-bisphosphate carboxylase/oxygenase,” but everyone knows it as “RuBisCo.” Its function in the cell is to take the carbon of carbon dioxide, obtained from the air, and link it to a five-carbon sugar. This makes a six carbon sugar, an essential part of the process of photosynthesis. But it also allows the carbon to be used in a variety of other chemical reactions inside a cell that would never work with carbon dioxide. These include creating the building blocks of DNA and proteins. Through these two functions, the enzyme is essential to most life on Earth.

It’s pretty easy to grow vast quantities of microbes. It’s a lot harder to convert those microbes into something useful. For example, we’ve engineered algae that, when starved for nitrogen, will put most of their stored energy into fats. Fats are chemically similar to hydrocarbons, so this is potentially useful for making biofuel. But “similar” isn’t really good enough; you still need to process the fats before they can be used as a fuel.

But some researchers may have figured out a way to get biology to convert fats directly to hydrocarbons using nothing more than sunlight. They’ve identified an enzyme that catalyzes the conversion of fats to long hydrocarbons, which could be used as fuel with no further modification.

Fat vs. fuel

Gasoline is a mixture of different hydrocarbons, which are molecules that contain only hydrogen and carbon. The simplest hydrocarbons are linear chains, with a backbone of carbon atoms where each carbon is linked to, at most, two others (with two or three hydrogen atoms also linked to each carbon). The chemical processing done in refineries ensures that gasoline mostly contains branched hydrocarbons, wherein some carbon atoms are linked to three or four other carbons.

The chemical enzyme is sent by fat cells as a sensor to indicate enough food has been eaten.

This stops any more eating – preventing us from putting on too much weight.

The breakthrough could could also lead to treatments for eating disorders such as anorexia nervosa.

Drugs that target the protein known as BH4 by boosting or lowering levels could suppress the appetite of those hooked on food or increase it for others addicted to dieting.

Experiments on flies found it plays a key role in reducing eating in flies – offering hope of a similar chemical pathway in humans.

Professor Walton Jones, of the Korea Advanced Institute of Science and Technology in Daejeon, said: “Our study indicates fat tissue sends a molecular signal to the fly brain to regulate feeding behaviour.

“Further studies will be needed to determine if a similar system acts in mammals, and if so, whether it can be safely manipulated to help achieve weight loss, or gain, in people.”

The study published in PLOS Biology said fat is the primary long-term energy storage chemical in animals and controlling levels is critical for survival.

In mammals the hormone leptin induces eating in response to fat loss but so far no corresponding signal has been identified stopping eating in response to fat gain.

Fruit flies replicate many of the feeding-related regulatory mechanisms and genes known to operate in humans.

Professor Jones, a neurobiologist, explained this is why they make a good model for the search for such an inhibitory signal.

His team focused on bits of DNA called microRNAs as they are well-known inhibitors of gene expression.

This identified the chemical miR-iab-4 in fat tissue which increased feeding by more than 27 per cent and a gene called purple which was expressed in fat bodies.

Blocking the gene increased feeding – suggesting its normal function was to inhibit it.

Purple is known to be one of two enzymes that build a molecule called PTP which is released by fat bodies and circulates in the fly brain.

There, a third enzyme converts PTP into BH4 (tetrahydrobiopterin) where neurons produce the protein NPF that regulates feeding.

Loss of purple in the fat body – or of BH4 in brain cells – led to increased release of NPF and increased feeding.

Conversely, increasing BH4 in neurons reduced NPF release and decreased feeding.

Finally, the researchers showed feeding flies a low-calorie diet reduced expression of the fat body enzymes that control BH4 production – and led to increased feeding.

They said the results suggest BH4 plays a key role in suppressing appetite in flies – and that PTP released from fat bodies delivers a signal to the brain indicating energy stores are sufficient and feeding can stop.

Professor Jones said: “Because the enzymes that produce BH4 and the enzymes that require BH4 are all highly conserved, it will be interesting to see whether our results in flies also apply to mice or humans.”
