He Jiankui (pictured) told the Associated Press that twin girls, Lulu and Nana, were born earlier this month following embryo-editing using CRISPR to disable the CCR5 gene, which allows the HIV virus to infect cells. An American scientist, Michael Deem, also reportedly assisted He on the project at the Southern University of Science and Technology of China.

According to He, embryos were edited for seven couples affected by HIV, with one pregnancy resulting thus far. He added that the patients refused to be identified or interviewed — he also wouldn’t say where the work was done.

TIP: A Guide To Genetic Engineering Biotech And How It Works

The news is already causing an outcry among researchers in the US, who slammed it as “unconscionable” and “immoral and unethical.” It follows similar reactions to China’s breakthrough on genetically-modified human embryos in 2015, which saw researchers remove a gene called HBB, responsible for the fatal blood disorder β-thalassaemia.

Notably, He’s work has not been published in a peer-reviewed scientific journal, meaning there’s no independent confirmation of the research. In an earlier report, the MIT Technology Review linked to Chinese medical documents posted online in the Chinese Clinical Trial Registry — a primary registry of the World Health Organization’s International Clinical Trial Registry. Scientists shown He’s materials by the AP concluded that the tests so far are insufficient to say the editing worked or to rule out harm to the genetically-edited babies.

Both the UK and Japan have given the green light to edit donated human embryos to better understand developmental processes. Meanwhile, the National Academy of Sciences also endorsed embryonic engineering last year, but only in cases that would otherwise result in children being born with serious genetic diseases.

  • Discord has shut down a chat group on its service that was being used to share pornographic videos that had been edited using an artificial intelligence technology to include the images of female celebrities without their consent.
  • Dozens of people were active on the channel, which Discord said violated its rules on revenge porn.
  • People who have doctored videos using the AI technology are also sharing them on Reddit.

Online chat service Discord has shut down a user-created chat group that was being used to share pornographic videos that had been doctored using artificial intelligence technology to include the images of female celebrities without their consent.

The company closed down the group shortly after Business Insider reached out to Discord about it. Discord took it offline because the chat group violated its rules against non-consensual pornography — otherwise known as revenge porn, a company representative said in a statement.

“Non-consensual pornography warrants an instant shut down on the servers whenever we identify it, as well as permanent ban on the users,” the representative said. “We have investigated these servers and shut them down immediately.”

Discord lets users create their own communities or “servers” around certain subjects. Originally intended as a tool for gamers to communicate, it is now used more broadly, with more than 14 million daily users it says, who send more than 300 million daily messages.

The chat group was called “deepfakes”, and it included several channels in which users could communicate. One was reserved for general discussions, another for sharing pictures that could be used to train the AI technology, and yet another for sharing the doctored videos. About 150 Discord users were logged into the deepfakes server earlier on Friday.

The users who created the chat group did attempt to set some ground rules for it. The rules stated that the videos couldn’t be doctored to include images of “amateurs;” instead, only images of celebrities and public figures were allowed. And the rules barred images or videos of children.

“It goes without saying that you should respect each other,” the rules added. “Be polite, avoid excessive swearing, and speak English at all times.”

Over the last few months, growing numbers of people have been using an AI technology called FakeApp to virtually insert images of celebrities and other people into videos in which they didn’t originally appear, as first reported by Motherboard in December 2017. People who are interested in the technology or who have already edited videos using it have been congregating on Discord and Reddit to exchange notes on how to use FakeApp and share their efforts. (A Reddit spokesperson did not respond to a request for comment.)

Although the technology can be used for a variety of purposes, to date it’s largely been used to place the images of famous women into adult videos without their consent.

As of publication, a Reddit community dedicated to sharing the clips remained online, and users were discussing the closure of the Discord channel. Some had been banned from all servers across the service, they said.

“The celeb faceswap gifs and videos seem to be slowly disappearing from this [subreddit], and now the discord is gone… I’m worried that the best fap fuel may be about to be buried,” one user wrote.

“Of course this has more potential than that, but come on. The porn is clearly the best part.”

A team of researchers in Oregon have become the first in the US to attempt genetically altering human embryos, according to reporting by MIT Technology Review. The attempt is said to represent an advance in the safety and efficacy of methods used to correct genetic defects that spur disease.

Until now, the only three published reports of human embryo gene editing were from researchers in China. But their experiments—using a gene-editing method called CRISPR—caused “off-target” genetic changes, basically sloppy changes in the DNA that were not intended. Also, not all the cells in the embryos were successfully edited, causing an effect called “mosaicism.” Together, the problems suggested that the technique was not advanced enough to safely alter human embryos without unintended or incomplete genetic consequences.

Scientists familiar with the new US work told MIT Technology Review that the Oregon team has improved these issues. They’re said to have shown in experiments with “many tens” of human embryos that they can correct genetic mutations that cause disease while avoiding mosaicism and off-target effects. Their improved method allows for earlier delivery of CRISPR into cells at the same time sperm fertilize an egg.

DNA of human embryos edited for 1st time in US – report https://briefnews.eu/dna-of-human-embryos-edited-for-1st-time-in-us-report/ https://briefnews.eu/dna-of-human-embryos-edited-for-1st-time-in-us-report/#respond Thu, 27 Jul 2017 04:17:43 +0000 http://briefnews.eu/dna-of-human-embryos-edited-for-1st-time-in-us-report/ A team of researchers in Oregon have become the first to attempt to create genetically modified human embryos in the United States. The team reportedly demonstrated they could eliminate diseases in offspring with CRISPR.

Led by Shoukhrat Mitalipov, a team of scientists at the Oregon Health and Science University used the CRISPR-Cas9 genome editing technology to alter human DNA in single-cell embryos, according to a report published Wednesday on the MIT Technology Review.

“So far as I know, this will be the first study reported in the US,” Jun Wu, a collaborator at the Salk Institute, told the MIT Technology Review.

The report claims that Mitalipov broke new ground “both in the number of embryos experimented upon and by demonstrating that it is possible to safely and efficiently correct defective genes that cause inherited diseases.”

The team used donated sperm from men who carried inherited disease mutations to demonstrate that the genes can be corrected. Although the report said that “many tens” of embryos were created in the experiment, they were never intended to be implanted into a womb and were only allowed to develop for a few days.

To eradicate the disease, Mitalipov and his team used a process called “germline engineering,” in which genetically modified organisms pass changes to their offspring.

Earlier publications by scientists in China found that CRISPR caused editing errors known as “mosaicism,” in which some cells in the embryo did not take the desired changes. However, Mitalipov said his team was able to avoid mosaicism by injecting CRISPR into the eggs at the same time they were fertilized with sperm.

The technique was previously tested by Tony Perry, who was able to successfully edit the genes in mice to change the expected fur color of their offspring.

A scientist familiar with Mitalipov’s project said the tests were “proof of principle that it can work.”

“They significantly reduced mosaicism. I don’t think it’s the start of clinical trials yet, but it does take it further than anyone has before,” an unnamed scientist told the MIT Technology Review.

Mitalipov did not confirm the results of the test, telling the MIT Technology Review that they are still “pending publication.” However, if his research passes peer review it could be a significant step for scientists in the US.

Successfully editing genes in human embryos could theoretically lead to scientists being able to cure any number of diseases, including cancer.

To date, there have only been three previous reports of editing human embryos, all of which were published by scientists in China.

Federal regulations have barred scientists in the US from any efforts to turn an editing embryo into a baby. Many critics are worried that the practice could lead to “designer babies” that are engineered with genetic enhancements.

Last year, the Office of the Director of National Intelligence released a report that added genome editing to a list of weapons of mass destruction and proliferation, saying it “increases the risk of the creation of potentially harmful biological agents or products.”

In February, the US National Academy of Sciences released a report, endorsing the use of gene editing, but only for “serious conditions under stringent oversight.”

Scientists Create the First Gene Edited, Tuberculosis-Resistant Cows https://briefnews.eu/scientists-create-the-first-gene-edited-tuberculosis-resistant-cows/ https://briefnews.eu/scientists-create-the-first-gene-edited-tuberculosis-resistant-cows/#respond Wed, 01 Feb 2017 15:29:59 +0000 http://briefnews.eu/scientists-create-the-first-gene-edited-tuberculosis-resistant-cows/

Gene editing research is moving quickly in China—researchers there have already edited human embryos, after all. But a team of scientists now have their sights set on the food supply. Ladies, gentelmen, etc., meet the first crop of tuberculosis resistant, genetically moo-dified cows.

Sorry.

Tuberculosis has long been a problem for cattle farmers in developing countries, especially in Asia and Africa. Now, scientists from Northwest A&F University in Shaanxi, China have used a tweaked CRISPR/Cas9 gene-editing system to insert a protein that helps fight off the disease-causing bacteria. The cows developed some resistance to tuberculosis (moo-berculosis?), though US Food and Drug Administration regulations may prevent us from seeing these cows in the US for a long time. This is the first time gene editing has been used to confer tuberculosis resistance in cows, making the study a major breakthrough.

CRISPR/Cas9 is the best gene editing system scientists have right now—in simple terms, it’s like scissors and a glue stick for DNA. Regular versions of the Cas9 enzyme breaks both strands of the DNA helix, which can cause unexpected changes in the DNA code, moo-tations if you will. In this study the researchers used a modified version of the enzyme, Cas9 nickase or Cas9n, which simply “nicks” a single strand and allows for more specific editing, and fewer harmful moo-tations, to insert a gene which helps provide infection resistance.

The authors cut the TB resistance-conferring gene, natural resistance-associated macrophage protein-1 or NRAMP1, from a line of mouse white blood cells that you can buy yourself online, and glued it into over a thousand cow embryos, which they transplanted into over four hundred cows. This resulted in 20 genetically modified cows, 11 that lived longer than three moo-nths (ok sorry I’m done).

When the researchers infected the cows with Mycobacterium bovis, which causes tuberculosis, their genetically modified creations showed more resistance than the non-gene edited cows. The researchers published their results in the journal Genome Biology today.

Giving cows bacterial resistance to diseases could be a huge deal. Cows are a major cause of the impending superbug crisis, since we feed them lots of antibiotics to prevent them from getting sick in filthy, overcrowded living situations. That has caused some bacteria to evolve antibiotic resistance and turn into so-called “superbugs,” which could kill millions of people annually if we don’t slow our antibiotics use. Humans are looking into some alternatives to antibiotics to deal with the crisis.

We probably won’t see these cows in the United States for a long time, however. Just a few weeks ago, the United States Food and Drug Administration introduced draft rules stating that gene-edited animals needed to be tested for safety and efficacy as if they were new drugs. However, Chuck Sattler, the vice president of genetic programs at Select Sires, told Newsweek that some cows already have tuberculosis resistance-conferring gene—meaning we might be able to breed these cows to get around the tough FDA regulations.

So anyway, we just thought you should know that we can’t have nice things like cool genetically modified disease-resistant cows here in the United States.
