Believe it or not, scientists are still discovering new forms of stem cells — and these latest examples could shake up how doctors treat a multitude of common injuries. A Stanford-led research team has identified the human skeletal stem cell, helping the group create a “family tree” of cells that can regenerate bones and cartilage. You can either isolate them from existing bones or generate them from specialized cells in fat, and they’re predictable enough (that is, they’ll always make bone tissue) that doctors wouldn’t have to worry much about unintended results when using them in treatments.

The breakthrough wasn’t a simple affair. To pinpoint the human skeletal stem cell, the scientists couldn’t just use the tricks they’d used to isolate the equivalent in mice. They had to compare the mouse’s gene expression profiles with those of several human cell types you’d find on the growing ends of human bone. That let the group find cells with similar proteins as the mouse’s skeletal stem cells, helping the team find relevant markers on human cells.

The findings will should help understand the nature of human bone, but Stanford noted that it’s ultimately interested in medical uses. You could heal broken bones at a faster pace, repair cartilage or even grow new bones for reconstructive surgery. Conditions like arthritis and osteoporosis might be much less serious, as you could generate unaffected bones and cartilage as necessary. This is only the beginning, so any solutions are likely years away. Still, there could be a day when you don’t have to worry as much about serious fractures or the effects of aging.

Citizen science, which asks the public to help out science projects, has produced some spectacular successes. But finding a way to grab and maintain hold of the public’s attention can be a challenge. That has led to a number of projects that turn the science challenge into a game, finding ways of making a “win” into scientific progress.

But scientists have also figured out ways of hijacking existing games, including using pre-existing fan bases that recruit players through in-game rewards. Now, there’s a progress report on an effort to turn EVE Online players into cell biology experts. Thanks to some in-game rewards, more than 300,000 players contributed roughly 33 million calls on where in a cell a protein was located. This not only greatly expanded a public database of information on proteins, but it enabled the researchers to better train a neural network to do the same thing.

Call it

While in many cases it has been possible to determine or infer what a protein does, that only gives us a partial idea of its actual function. That’s because many proteins are shipped to specific locations in cells. So while two proteins may look similar in terms of the order and identity of their amino acids, one may be shipped to the nucleus, where it interacts with DNA, while its relative gets sent to the cell’s surface, where it acts on proteins in the surroundings. So figuring out where a protein normally resides within cells can go a long way toward helping us figure out its normal functions.

Over the course of their careers, biologists develop a huge mental library of cell structures and their corresponding data. Investigating specific areas of a living cell involves a piecemeal approach, identifying how some parts work with others and spending time on cell labelling. But now, the Allen Institute for Cell Science has launched the first predictive 3D model of a live human cell — the Allen Integrated Cell — and it could be “a total game changer”, according to researchers.

“This is a new way to see inside living human cells,” said Rick Horwitz, executive director of the Allen Institute for Cell Science. “It’s like seeing the whole cell for the first time. In the future, this will impact drug discovery, disease research and how we frame basic studies involving human cells.”

To create the breakthrough model, scientists gene edited a large collection of live human cells to incorporate fluorescent protein tags, which illuminate specific structures inside the cells. The team took tens of thousands of pictures of these glowing cells, and used AI to create a probabilistic model, which predicts the most probable shape and location of structures in any cell, based on the shape of the plasma membrane and the nucleus.

The team then applied a different machine-learning algorithm to those pictures, which used what it learned from cells with fluorescent labels to find cellular structures in cells without fluorescent labels. By combining all the data, the system was able to generate images that look nearly identical to those obtained by traditional fluorescence microscopy, which can be expensive and toxic.

“Until now, our ability to see what is going on inside of human cells has been very limited,” said Michael Elowitz, professor of biology, bioengineering and applied physics at California Institute of Technology. “Previously, we could only see the proteins that we deliberately labeled. But the Allen Integrated Cell is like the ultimate free lunch. We get to sample a ‘buffet’ of many different proteins and organelles, without having to label anything at all. This opens up a totally new and much more powerful way of doing cell biology. It’s a total game changer.”

The premiere of the second season of Westworld is a perfect time to ponder what makes us human. This is not new territory; such questions have long been dealt with in works of fiction, and they have appeared in science in the form of studies of creatures that have human-like characteristics—like consciousness—yet are not Homo sapiens.

These studies raise ethical questions whether the subject is an animal or an AI. Last May, a consortium of bioethicists, lawyers, neuroscientists, geneticists, philosophers, and psychiatrists convened at Duke to discuss how this question may apply to relatively new entities: brain “organoids” grown in a lab. These organoids can be either chimaera of human or animal cells or slices of human brain tissue. Will these lab-grown constructs achieve any sort of consciousness deserving of protection?

Why organoids?

If we ever want to understand, let alone cure, the very complex brain disorders that plague people—like schizophrenia, ASD, and depression—we need research models. And in order to be informative, these models must be accurate representations of the human brain. Yet as our models become more and more like the real thing (and for now, they are still quite a long way off), the problems with using them become so pronounced as to negate their utility—like Borges’ map.

Norovirus inflames the stomach and/or intestines and causes pain, nausea, vomiting, and diarrhea. It is super contagious and kills tens of thousands of people each year. But until now, we did not even know which cells it targets to create all this havoc. A recent study by a public-private consortium working in universities and Genetech has just discovered the elusive cell type (in mice): they’re called tuft cells, and they reside in the ilium and colon.

Obviously, norovirus attacks intestinal epithelial cells, the specialized cells that line tubes within the body. But last year, the same group reported that noroviruses would infect only a rare subset of them and not most of their neighbors. But the researchers could not discern what made these select cells so special.

They knew that norovirus used a particular receptor to infect cells and that this receptor is both necessary and sufficient for infection. Oddly, the receptor is an immunoregulator thought to be expressed by blood-forming cells, specifically immune progenitor cells in the bone marrow. These could make their way to the intestines once they mature. But mice that got bone marrow transplants that lacked this receptor were still susceptible to norovirus infection, so that’s clearly not the case.
